Dialysis
- Precipitate NH4SO4 using with 40mM, 50mM, 60mM, 70mM, 80mM to optimize the enzyme activity of protein,
- MOPs/NaOH 20mM, pH.7 usually, 150 times-volume of solution to be dialyzed
- Dialyze overnight
The reasons to precipitation and dialysis are to eliminate low Molecular weight protein, and NH4SO4 that keep protein unactive.
Extract pig heart put in the
Test enzyme activity:
Crude extract gives 0.033 units/min
After dialysis, it gives 0.005 units/min
Then  the anion exchange chromatography, The fractions are collected and test enzyme activity.
There are 2 fractions showing high enzyme activity at 0.011 and 0.008 units/min. Continue with SDS-PAGE(Run electrophoresis)
